Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.

Regulatory treatment varies between countries. Several national medicines agencies have classified the peptide as unapproved, and customs authorities in some jurisdictions seize shipments on that basis. A few jurisdictions channel supply through prescription-only frameworks that do not list the substance by name. Because the material circulates mainly through online vendors, composition and purity are rarely verified before sale. Surveys of unapproved peptide products have reported labels that did not match measured content in a substantial fraction of samples.
Melanotan II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its structure substitutes a lactam bridge between side chains to increase stability relative to the native hormone. The compound is also known by the shorthand MT-II and by several non-proprietary synonyms used in research catalogues. It is not an approved therapeutic product in any major jurisdiction; material sold under this name is typically offered as a laboratory reagent rather than as a medicine.
The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.

Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Chapter 8 of the 1977 Soviet Constitution is titled as the "Soviet Union is a union state". Article 70 stated that the union was founded on the principles of "socialist federalism" as a result of the free self-determination of nations and the voluntary association of equal Soviet Socialist Republics. Article 71 listed all fifteen union republics that united into the Soviet Union. According to Article 76 of the 1977 Soviet Constitution, a union republic was defined as a sovereign Soviet socialist state that had united with other Soviet Republics into the USSR. Article 78 of the Constitution stated that the territory of a union republic may not be altered without its consent. The boundaries between republics may be altered by mutual agreement of the republics concerned, if the rest of the union agreed. Article 81 of the Constitution stated that "the sovereign rights of Union Republics shall be safeguarded by the USSR". In the final decades of its existence, the Soviet Union officially consisted of fifteen Soviet Socialist Republics (SSRs). All of them, with the exception of the Russian SFSR (until 1990), had their own local party chapters of the All-Union Communist Party. In 1944, amendments to the All-Union Constitution allowed for separate branches of the Red Army for each Soviet Republic. They also allowed for Republic-level commissariats for foreign affairs and defense, allowing them to be recognized as de jure independent states in international law.
In the proposed biosynthetic pathway, Scheme 1, L-Phe (1) is used as the starting material. It is converted into Cinnamic acid (2) via phenylalanine ammonia lyase (PAL). Then it is turned into p-Coumaric acid (3) with use of cinnamate 4-hydroxylase (C4H). 4-coumarate:CoA ligase (4CL) is then used to get p-Coumaroyl-CoA (5). P-Coumaroyl shikimate transferase (CST) is the enzyme that is responsible for the bonding of shikimic acid and p-Coumaroyl-CoA. The complexed (5) is then selectively oxidized at C3 by p-coumaroyl 5-O-shikimate 3'-hydroxylase (CS3'H) to alcohol. With another action of CST, shikimate is broken off from this intermediate, thereby yielding Caffeoyl-CoA (7). In order to get desired substitution pattern on the aromatic ring, caffeoyl-CoA O-methyltransferase (CCOMT) converts the hydroxyl group at C3 into methoxy as seen in Feruloyl-CoA (8). Up until this step, according to Ramirez-Ahumada et al., the enzyme activities are very active. It is speculated that some polyketide synthases (PKS) and reductases are involved in final synthesis of 6-Gingerol (10).
The attacks had a significant economic impact on the U.S. and world markets. The stock exchanges did not open on September 11 and remained closed until September 17. Reopening, the Dow Jones Industrial Average (DJIA) fell 684 points, or 7.1%, to 8921, a record-setting one-day point decline. By the end of the week, the DJIA had fallen 1,369.7 points (14.3%), at the time its largest one-week point drop in history. In 2001 dollars, U.S. stocks lost US$1.4 trillion in valuation for the week. In New York City, about 430,000 job months and US$2.8 billion in wages were lost in the first three months after the attacks. The economic effects were mainly on the economy's export sectors. The city's GDP was estimated to have declined by US$27.3 billion for the last three months of 2001 and all of 2002. The U.S. government provided US$11.2 billion in immediate assistance to the Government of New York City in September 2001, and US$10.5 billion in early 2002 for economic development and infrastructure needs. Also hurt were small businesses in Lower Manhattan near the World Trade Center (18,000 of which were destroyed or displaced), resulting in lost jobs and wages. Assistance was provided by Small Business Administration loans; federal government Community Development Block Grants; and Economic Injury Disaster Loans. Some 31,900,000 square feet (2,960,000 m2) of Lower Manhattan office space was damaged or destroyed. Many wondered whether these jobs would return, and if the damaged tax base would recover.
In 1993, Jeffrey Ulmer and co-workers at Merck Research Laboratories demonstrated that direct injection of mice with plasmid DNA encoding a flu antigen protected the animals against subsequent experimental infection with influenza virus. In 2016 a DNA vaccine for the Zika virus began testing in humans at the National Institutes of Health. The study was planned to involve up to 120 subjects aged between 18 and 35. Separately, Inovio Pharmaceuticals and GeneOne Life Science began tests of a different DNA vaccine against Zika in Miami. The NIH vaccine is injected into the upper arm under high pressure. Manufacturing the vaccines in volume remained unsolved as of August 2016. Clinical trials for DNA vaccines to prevent HIV are underway. In August 2021, Indian authorities gave emergency approval to ZyCoV-D. Developed by Cadila Healthcare, it is the first DNA vaccine against COVID-19.

=== Pregnancy === Antidepressant exposure (including escitalopram) is associated with shorter duration of pregnancy (by three days), increased risk of preterm delivery (by 55%), lower birth weight (by 75 g), and lower Apgar scores (by <0.4 points). Antidepressant exposure is not associated with an increased risk of spontaneous abortion. There is a tentative association of SSRI use during pregnancy with heart problems in the baby. The advantages of their use during pregnancy may thus not outweigh the possible negative effects on the baby.
== Research == A synthetic antibody has been shown to neutralize a major class of neurotoxins produced by four deadly snake species from South Asia, Southeast Asia, and Africa. The antibody targets long-chain α-neurotoxins, a common and lethal component of many elapid venoms, and may represent a step toward a universal antivenom effective against a broad spectrum of snake species. Separately, a combination of broadly neutralizing human antibodies and the phospholipase inhibitor varespladib has been shown to protect mice from venom-induced lethality caused by multiple snake species. The antibodies were derived from a hyperimmune human donor who had developed broad resistance to snake venom through repeated exposures, resulting in a unique immune profile capable of neutralizing diverse venom toxins.
"(...) Chile's position towards the Peru-Bolivian Confederation is untenable. It cannot be tolerated either by the people or by the Government because it is tantamount to their suicide. We cannot look without concern and the greatest alarm, the existence of two peoples, and that, in the long run, due to the community of origin, language, habits, religion, ideas, customs, will form, as is natural, a single nucleus. United these two States, even if it is only momentarily, will always be more than Chile in every order of issues and circumstances (...) The confederation must disappear forever and ever from the American scene due to its geographical extension; for its larger white population; for the joint riches of Peru and Bolivia, scarcely exploited now; for the dominance that the new organization would try to exercise in the Pacific by taking it away from us; by the greater number of enlightened white people, closely linked to the families of Spanish influence that are in Lima; for the greater intelligence of its public men, although of less character than the Chileans; For all these reasons, the Confederation would drown Chile before very soon (...) The naval forces must operate before the military, delivering decisive blows. We must dominate forever in the Pacific: this must be their maxim now, and hopefully it would be Chile's forever (...)”.
=== Coordinated healing === DeMarco’s animal studies showed a healing phenomenon he termed "coordinated healing". Observing that animal wounds would heal differently when treated with his formula, he tried to explain his observations: after excising a square inch of skin from a rabbit’s shaved back, he would allow it to heal normally while documenting its progress with time-lapse photography. DeMarco then performed the same experiment while treating the rabbit with his formula. He observed that the untreated wound healed haphazardly; closing in a non-circular pattern with uneven levels and with infiltrating hair growth before wound closure. In contrast, the treated rabbit shows healing in a smooth circular pattern with even levels and only after wound closure would the hair begin to regrow.
=== Partner proteins === Assembly of the eIF4F complex is inhibited by proteins known as eIF4E-binding proteins (4E-BPs), which are small heat-stable proteins that block cap-dependent translation. Non-phosphorylated 4E-BPs interact strongly with eIF4E thereby preventing translation; whereas phosphorylated 4E-BPs bind weakly to eIF4E and thus do not interfere with the process of translation. Furthermore, binding of the 4E-BPs inhibits phosphorylation of Ser209 on eIF4E. Of note, 4E-BP1 is found in both the nucleus and the cytoplasm, indicating that it likely modulates nuclear eIF4Es functions of eIF4E as well. A recent study showed that 4E-BP3 regulated eIF4E dependent mRNA nucleo-cytoplasmic export. There are also many cytoplasmic regulators of eIF4E that bind to the same site as 4E-BP1. Many other partner proteins has been found that can both stimulate or repress eIF4E activity, such as homeodomain containing proteins, including HoxA9, Hex/PRH, Hox 11, Bicoid, Emx-2 and Engrailed 2. While HoxA9 promotes mRNA export and translation activities of eIF4E, Hex/PRH inhibits nuclear functions of eIF4E. The RNA helicase DDX3 directly binds with eIF4E, modulates translation, and has potential functions in P-bodies and mRNA export. RING domains also bind eIF4E. The promyelocytic leukemia protein PML is a potent suppressor of both the nuclear RNA export and oncogenic activities of eIF4E whereby the RING domain of PML directly binds eIF4E on its dorsal surface suppressing eIF4E's oncogenic activity; and moreover a subset of PML and eIF4E nuclear bodies co-localize.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
It is a synthetic cyclic heptapeptide and an analogue of alpha-melanocyte-stimulating hormone. The molecule is produced by chemical synthesis rather than extracted from a biological source.
