Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanocortin receptors comprise five subtypes with distinct tissue distributions and functions. Melanotan-2 is described in the literature as a non-selective agonist that engages several of these subtypes, including MC1R, MC3R, MC4R, and MC5R. MC1R is the subtype most directly linked to melanin production in skin cells. Because the compound is not subtype-selective, its observed effects in experimental settings are generally attributed to activity across multiple receptor pathways rather than to a single target.
Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.

Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
==== Cylindrical spirals myopathy ==== Cylindrical spirals myopathy is very rare with only 18 individual cases described as of 2013. The majority of cases are sporadic, and has been observed in only 3 families. It is characterized by the presence of cylindrical spirals as the main pathological finding in muscle biopsies. Cylindrical spirals are unusual membrane structures that have a spiral pattern. These membrane structures are seen during electron microscopic examination of the affected muscle. These structures merge into or are surrounded by tubular structures that resemble tubular aggregates. Tubular aggregates are abnormal accumulations of membranous tubules and have been observed in a wide variety of muscle diseases and originate from the sarcoplasmic reticulum. Cylindrical spirals were first described in 1979 and were thought to be a non-specific reaction of skeletal muscle secondary to a metabolic disturbance or muscle fibre injury. The molecular basis of cylindrical spiral myopathy is currently unknown, however a genetic mutation affecting the sarcoplasmic reticulum in some patients seems likely, as SERCA1, calsequestrin, and RYR1 have been shown to bind to cylindrical spirals. Cylindrical spirals have also been shown to react with the mitochondrial enzyme succinate dehydrogenase, which suggests that cylindrical spirals originate from mitochondria. Phenotypes are quite variable, and manifestations can include weakness, abnormal gait, myotonia, cramps, and scoliosis.
As in education, there is a universal human right to 'health and well-being' including 'medical care and necessary social services', although human rights law does not say what the best system of health governance is. Among EU member states there are two main traditions of health care provision, based on public service or insurance. First, healthcare may be seen as a public service free at the point of use, with hospitals largely owned by the public health service and doctors publicly funded (the "Beveridge model".) This is the system, for example, in Finland, Sweden, Denmark, Spain, Italy, Portugal, Greece or Ireland. Second, healthcare can be provided through insurance, where hospitals and doctors are separately owned and run from the service provider (the "Bismarck model"). There is a large spectrum between systems based mainly on public insurance and usually creating public option hospitals or requiring no profit (such as France, Belgium, Luxembourg, Slovenia, the Czech Republic or Estonia) and those that allow substantial private and profit-making insurance and hospital or doctor provision (the Netherlands and Germany). In all cases, health is universal, and subsidised or free wherever people cannot afford insurance premiums. The healthcare outcomes vary greatly between different systems, so that while there is generally higher life expectancy with more investment, healthcare tends to have worse outcomes and costs more where there is more private business or profit.
On 20 January, the Brazilian Ministry of Health declared a national emergency following reports of deaths among Yamomami children due to malnutrition and easily curable diseases. The president of the FUNAI also led the creation of a task force to deal with the humanitarian crisis. Between 2019 and 2023, at least of 570 Yanomami children died from malnutrition, hunger and mercury contamination. Lula accused Jair Bolsonaro's government of having committed genocide against the Yanomami, several researchers blamed the entry of illegal miners into protected native lands for the high amount of deaths between the local indigenous peoples, as well as alleged connivance and omission of the federal and local/state administrations during the previous government. Due to the high number of deaths since 2023, reports on the situation decreased, until they stopped being published at the beginning of 2024, when there was a further increase in deaths due to violence.
In February 2020, bempedoic acid was approved for use in the United States both as a standalone drug (brand name Nexletol) and in a fixed-dose combination with ezetimibe (brand name Nexlizet). The U.S. Food and Drug Administration (FDA) granted the approval of Nexletol to Esperion Therapeutics. The FDA approved bempedoic acid based on evidence from two clinical trials (Trial 1/ NCT02666664 and Trial 2/NCT02991118) of 3009 subjects with high LDL cholesterol and known atherosclerotic cardiovascular disease or HeFH. The trials were conducted in United States, Canada, and Europe.
==== Effects of pH ==== The uranium fraction diagrams in the presence of carbonate illustrate this further: when the pH of a uranium(VI) solution increases, the uranium is converted to a hydrated uranium oxide hydroxide; At high pH, it becomes an anionic hydroxide complex. When carbonate is added, uranium is converted to a series of carbonate complexes if the pH is increased. One effect of these reactions is increased solubility of uranium in the pH range 6 to 8, a fact that has a direct bearing on the long term stability of spent uranium dioxide nuclear fuels.

[W]e find that the European Communities did not actually proceed to an assessment, within the meaning of Articles 5.1 and 5.2, of the risks arising from the failure of observance of good veterinary practice combined with problems of control of the use of hormones for growth promotion purposes. The absence of such risk assessment, when considered in conjunction with the conclusion actually reached by most, if not all, of the scientific studies relating to the other aspects of risk noted earlier, leads us to the conclusion that no risk assessment that reasonably supports or warrants the import prohibition embodied in the EC Directives was furnished to the Panel. We affirm, therefore, the ultimate conclusions of the Panel that the EC import prohibition is not based on a risk assessment within the meaning of Articles 5.1 and 5.2 of the SPS Agreement and is, therefore, inconsistent with the requirements of Article 5.1.
The club, like most other organizations in Germany, including sports and football clubs, was dissolved by the occupying Allied authorities in the aftermath of the war. Club members reconstituted the team in 1946 as SG Probstheida under the auspices of the occupying Soviets. After playing as BSG Erich Zeigner Probstheida and then BSG Einheit Ost, the club merged with sports club SC Rotation Leipzig in 1954 and played in the DDR-Oberliga, East Germany's top-flight league, but earned only mediocre results. In 1963, the city of Leipzig's two most important sports clubs, SC Rotation and SC Lokomotive Leipzig, were merged, resulting in the founding of two new sides: SC Leipzig and BSG Chemie Leipzig.
It was first synthesized in 1540 by Valerius Cordus, who noted some of its medicinal properties. He called it oleum dulce vitrioli, a name that reflects the fact that it is synthesized by distilling a mixture of ethanol and sulfuric acid (known at that time as oil of vitriol). August Sigmund Frobenius gave the name Spiritus Vini Æthereus to the substance in 1730.
==== Public figure ==== Throughout the fifties and sixties, Adorno became a public figure, not simply through his books and essays but also through his appearances in radio and newspapers. In talks, interviews, and round-table discussions broadcast on Hessen Radio, South-West Radio, and Radio Bremen, Adorno discussed topics as diverse as "The Administered World" (September 1950), "What is the Meaning of 'Working Through the Past?'" (February 1960), and "The Teaching Profession and its Taboos" (August 1965). Additionally, he frequently wrote for Frankfurter Allgemeine, Frankfurter Rundschau, and the weekly Die Zeit. At the invitation of Wolfgang Steinecke, Adorno took part in the Darmstadt Summer Courses for New Music in Kranichstein from 1951 to 1958. Yet conflicts between the so-called Darmstadt school, which included composers like Pierre Boulez, Karlheinz Stockhausen, Luigi Nono, Bruno Maderna, Karel Goeyvaerts, Luciano Berio, and Gottfried Michael Koenig, soon arose, receiving explicit expression in Adorno's 1954 lecture, "The Aging of the New Music," where he argued that atonality's freedom was being restricted to serialism in much the same way as it was once restricted by twelve-tone technique. With his friend Eduard Steuermann, Adorno feared that music was being sacrificed to stubborn rationalization. During this time, Adorno not only produced a significant series of notes on Beethoven (which was never completed and only published posthumously) but also published Mahler: A Musical Physiognomy in 1960.

=== Microbial metabolites === Microbes compete for living space and nutrients. To survive in these conditions, many microbes have developed abilities to prevent competing species from proliferating. Microbes are the main source of antimicrobial drugs. Streptomyces isolates have been such a valuable source of antibiotics, that they have been called medicinal molds. The classic example of an antibiotic discovered as a defense mechanism against another microbe is penicillin in bacterial cultures contaminated by Penicillium fungi in 1928.
The outer membrane of Gram-negative bacteria contains lipopolysaccharide, which acts as a barrier to vancomycin penetration. This barrier is why vancomycin is mainly used to treat infections caused by Gram-positive bacteria (except some nongonococcal species of Neisseria). The large hydrophilic molecule of vancomycin is able to form hydrogen bond interactions with the terminal D-alanyl-D-alanine moieties of the NAM/NAG-peptides. Under normal circumstances, this is a five-point interaction. This binding of vancomycin to the D-Ala-D-Ala prevents cell wall synthesis of the long polymers of N-acetylmuramic acid (NAM) and N-acetylglucosamine (NAG) that form the backbone strands of the bacterial cell wall, and prevents the backbone polymers from cross-linking with each other.
A pterygium (pl.: pterygia or pterygiums) is any wing-like triangular membrane occurring in the neck, eyes, knees, elbows, ankles or digits. The term comes from the Greek word pterygion meaning "wing".

Afamelanotide is a linear analog that has received approval in some jurisdictions for a specific photoprotection indication. Melanotan II is cyclic and less selective across melanocortin receptors. The two are related but are not interchangeable in regulatory or clinical terms.
A compound that activates several receptor subtypes may produce effects beyond pigmentation. Selectivity influences which tissues respond and shapes the side-effect profile described in reports. Designing subtype-selective analogs is a stated goal of melanocortin research.
No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
